Sclareol có độ bay hơi rất thấp nên chỉ xuất hiện ở tỉ lệ nhỏ khi phân tích bằng GC. Phân tích HPLC cho thấy absolute clary sage thực tế chứa 70 đến 75 phần trăm sclareol và 13-epi-sclareol. Dữ liệu cho thấy hợp chất tự nhiên an toàn ở nồng độ thông thường.
Trên da. Dạng nguyên chất gây kích ứng trên da thỏ, ở 3 phần trăm thì không. Ở 10 phần trăm trên 25 tình nguyện không kích ứng. Ba mẫu 10 phần trăm thử trên 106 người, không có kích ứng. Một mẫu cho phản ứng mẫn cảm 1 trên 29 và 3 trên 26, hai mẫu còn lại không phản ứng. Trong human repeat insult patch test ở 3 phần trăm trên 35 và 39 tình nguyện không gây mẫn cảm.
Đường uống. LD50 cấp tính trên chuột vượt 5 g/kg. LD50 da trên thỏ vượt 5 g/kg.
Bán mãn tính. Cho chuột uống 8.8 mg/kg/ngày sclareol trong 30 ngày chỉ thấy độc tính nhẹ. NOAEL chuột là 8.8 mg/kg/ngày.
Ung thư. Sclareol có hoạt tính độc tế bào mạnh với nhiều dòng tế bào ung thư người và động vật, gây apoptosis. 13-epi-Sclareol ức chế tế bào ung thư vú và tử cung mạnh hơn Tamoxifen nhưng không độc với tế bào bình thường.
Sclareol has extremely low volatility, so it appears only as a small percentage when analyzed by GC. HPLC analysis shows that clary sage absolute typically contains 70 to 75 percent sclareol and 13-epi-sclareol. The natural compound is safe at standard concentrations.
Skin. Undiluted material was irritating to rabbit skin, while 3 percent was not. At 10 percent on 25 volunteers it was not irritating. Three 10 percent samples tested on 106 individuals showed no irritation. One sample produced 1 of 29 and 3 of 26 sensitization reactions, while the other two samples produced none. In human repeat insult patch tests at 3 percent on 35 and 39 volunteers it was not sensitizing.
Oral. Acute oral LD50 in rats exceeded 5 g/kg. Acute dermal LD50 in rabbits exceeded 5 g/kg.
Subchronic. Rats given 8.8 mg/kg/day sclareol by gavage for 30 days showed only marginal toxicity. The rat NOAEL is 8.8 mg/kg/day.
Cancer. Sclareol shows strong cytotoxicity against many human and animal cancer cell lines, inducing apoptosis. 13-epi-Sclareol inhibits breast and uterine cancer cells more potently than Tamoxifen while sparing normal cells.
