α-Pulegone xuất hiện ở lượng nhỏ trong buchu chemotype pulegone và một số tinh dầu khác. Đồng phân β-pulegone là dạng độc hơn và phổ biến hơn trong tinh dầu pennyroyal.

Độc cấp. Trên chuột nhắt đực tiêm phúc mạc 400, 500 hoặc 600 mg/kg α-pulegone gây độc gan và tử vong tỉ lệ thuận với liều. Ở 400 mg/kg không có chuột chết, nhưng ở cùng liều của (1R)-(+)-β-pulegone, 9 trên 16 chuột chết.

Cảnh báo. Do cấu trúc tương tự, α-pulegone được giả định có rủi ro tương tự β-pulegone, dù mức độ thấp hơn. Tinh dầu chứa pulegone nên tránh trong thai kỳ và liều uống cao.

α-Pulegone occurs at low levels in buchu (pulegone chemotype) and a few other oils. The β-pulegone isomer is the more toxic form and is more abundant in pennyroyal oils.

Acute. In male mice given α-pulegone at 400, 500 or 600 mg/kg intraperitoneally, dose-dependent hepatotoxicity and lethality were observed. At 400 mg/kg no mice died, while at the same dose of (1R)-(+)-β-pulegone, 9 of 16 mice died.

Caution. Given structural similarity, α-pulegone is presumed to share the risk profile of β-pulegone, though at a lower level. Pulegone-containing oils should be avoided in pregnancy and at high oral doses.

Trong các tài liệu tham khảo, (R)-(Z)-α-pulegone và (S)-(E)-α-pulegone được phân biệt và quan sát thấy ở các tinh dầu khác nhau. β-Pulegone là cấu tử chính của tinh dầu pennyroyal và đã có ghi nhận ngộ độc gan nặng và tử vong trên người do uống tinh dầu pennyroyal.

Chất chuyển hoá menthofuran còn độc hơn pulegone đối với gan. Phòng ngừa cộng dồn rủi ro cho tổng pulegone và menthofuran.

Reference literature distinguishes (R)-(Z)-α-pulegone and (S)-(E)-α-pulegone, observed in different oils. β-Pulegone is the principal constituent of pennyroyal oil and has been associated with severe hepatic poisoning and death in humans following pennyroyal ingestion.

The metabolite menthofuran is more hepatotoxic than pulegone itself. Total pulegone plus menthofuran is the practical risk parameter.
